Merck

FDA approves enlicitide, bringing an oral PCSK9 macrocycle to market

FDA approved LIPFENDRA, or enlicitide, on July 15, with Merck announcing the decision on July 16. The once-daily 20 mg oral macrocyclic PCSK9 inhibitor is indicated alongside diet and exercise to reduce LDL-C in adults with hypercholesterolemia, including HeFH. Its approved lipid indication should not be conflated with product-specific proof of cardiovascular-event reduction.

ACS Nano

Nanopore-Mediated Assembly Enables Precise and Continuous Synthesis of mRNA-Encapsulated Lipid Nanoparticles for COVID-19 Vaccines

A hollow-fiber membrane platform uses dense nanopore arrays to control localized mixing during lipid nanoparticle assembly. The continuous process generated tunable particle sizes, narrow distributions, and high encapsulation, with particle properties linked to membrane pore size. Multiple lipid chemistries and nucleic acid payloads were compatible, and cell and animal studies supported activity. Commercial GMP performance remains to be established.

ACS Applied Materials & Interfaces

Stabilizing Anionic mRNA Lipid Nanoparticles by Cleavable Crosslinking of Cholesterol

Acid-cleavable cholesterol-derived crosslinkers and PEG-diamines stabilized anionic LNPs that otherwise lost activity during purification. By tuning crosslinking architecture, researchers balanced structural integrity with RNA release and improved splenic mRNA expression. Mechanistic analyses implicated intracellular processing as well as release behavior. These formulation-specific animal results do not establish general T-cell targeting or clinical manufacturing readiness.

Setting expectations for current and future directions of native mass spectrometry in the biopharmaceutical industry

This industry perspective examines native mass spectrometry as a complement to established biopharmaceutical characterization methods. It discusses intact assemblies, binding, stoichiometry, and heterogeneity, while identifying throughput, sensitivity, automation, and access limitations. The authors emphasize integration with analytical workflows rather than universal replacement of existing methods.

Tempest Therapeutics

Tempest outlines targeted mRNA/LNP in vivo CAR-T program and first-trial plan

Tempest outlined TPST-4003, an in vivo CAR-T candidate using CD7-targeted mRNA/LNP delivery of a dual CD19/BCMA CAR. A planned investigator-initiated study would enroll approximately ten patients with nervous-system autoimmune diseases, initially including myasthenia gravis and multiple sclerosis, with dosing anticipated in the fourth quarter. This is a development plan, not human efficacy evidence for the in vivo candidate.

Hansoh Pharma

Hansoh licenses ex-Greater-China HS-20118 rights to Avere alongside proposed NextCure merger

Hansoh granted Avere exclusive development, manufacturing, and commercialization rights to HS-20118, also called AVR-001, outside Greater China. The oral cyclic IL-23R antagonist agreement includes $120 million in upfront payments, up to $2.18 billion in milestones, and royalties. Avere also announced a proposed NextCure merger. Conditional milestone value and a signed merger agreement do not represent realized payments or closing.

Optimized Lipid Nanoparticles with Tail‐Modified Ionizable Lipids for Safer mRNA Delivery

A library of 56 ionizable lipids combined biodegradable disulfide linkages with different hydrophobic tail lengths and branching patterns. Structure-activity testing linked tail architecture to endosomal escape, translation, and biocompatibility. Optimized particles reduced cytotoxicity and cytokine induction, with immune-response evaluation extending to a nonhuman primate model. These findings support lipid design, rather than establishing human safety.

Angewandte Chemie International Edition

Ligand‐Mediated Reprogramming Redirects Liver‐Tropic Ionizable Lipid Nanoparticles for Lung‐Selective mRNA Delivery

Starting with a liver-tropic lead from 90 degradable lipidoids, researchers attached small-molecule ligands to redirect delivery toward the lung. The modified particles showed increased vitronectin association and enhanced pulmonary mRNA expression, with genome-editing proof of concept. This modular chemical approach retains the parent scaffold, but its targeting performance remains demonstrated in preclinical experimental conditions.

Vilya Research / arXiv (preprint)

Vilya-1 preprint explores macrocycle conformations and permeability modeling

Vilya’s technical preprint describes an all-atom model for conformational sampling and developability prediction across macrocycles with diverse chemistries and noncanonical residues. The framework supports permeability modeling and molecular design within the company’s oral-macrocycle strategy. Computational benchmark performance does not demonstrate human oral bioavailability, efficacy, or safety; the report is a preprint rather than peer-reviewed clinical evidence.

Salubris

Salubris receives trial clearance for cyclic peptide SAL0167 in hyperlipidemia

Salubris announced NMPA clinical-trial clearance for SAL0167 tablets for hyperlipidemia and identified the proprietary candidate as a cyclic peptide. The authorization permits clinical development but does not establish human dosing, quantitative oral exposure, or lipid-lowering efficacy. General statements about cyclic-peptide advantages should not be treated as demonstrated properties of this particular candidate.

GEMMABio / PR Newswire

GEMMABio receives ARPA-H support for mRNA/LNP base-editing development

GEMMABio announced ARPA-H funding for RAPID, a collaboration with Profluent under the THRIVE program. The project aims to develop four LNP-delivered, mRNA-encoded base-editing products for selected founder mutations causing homozygous familial hypercholesterolemia or maple syrup urine disease. The release did not disclose an award amount. Editor selection and delivery development remain research objectives, rather than demonstrated patient benefit.

Pulmonary mRNA-LNP vaccines for rapid and durable protection against bacterial infection

Researchers designed a pulmonary mRNA-LNP vaccine that combines early innate immune activation with later antigen-specific protection. Intratracheal vaccination primed neutrophils and macrophages and subsequently generated adaptive responses against Pseudomonas aeruginosa, including drug-resistant isolates. Single-cell profiling supported a coordinated immune mechanism. The protection experiments were conducted in female mice, rather than patients.
